it is the scene when Kirk and McCoy are in the Klingon court being tried for the attack on a Klingon ship where the Chancellor was on. General Chang asks McCoy, "state your current medical status." McCoy responds with, "aside from a bit of arithritis, pretty good." the Klingons that are in the courtroom laugh. Chang then says, "you have a singular wit, doctor."

what does a "singular wit" mean?

Singular means "one" or "individual", so I believe he was saying McCoy had a unique or one-of-a-kind sense of humor.

That's what it means. McCoy had his own sense of humor that went with his personality. His humor, like all Star Trek humor, was a little on the dry side.
