In the episode called "Darkstar Rising" Magister GILHIL said that he himself is in charge of over 300 inhabited planets, does that mean there are Plumbers ordered out and stationed on 300+ planets.
Yeah, pretty much. They're like intergalactic police.

I'm pretty sure there are a lot more guys like GILHIL that command hundreds of planets. So yeah, there might be plumbers on thousands of planets. The universe is a pretty big place.

Well here's my take on the Plumbers..
Plumbers are in fact intergalactic police/troubleshooters who basically arrest and contain criminals in intergalactic prisons (this would also include the Null void dimension (with the aid of a Null void generator to open a door to said null void)
From what I've been able to gleen, There's usually at least 1 if not more plumbers on a planet (think of each planet as a small city or a large one with its own police force).
The Plumbers are there to stop criminal activity (most notably having creatures bringing "High Level" Tech to "Low Level" backwater Planets that shouldn't have this tech (Earth is a class 2 technology world, and any item brought onto the planet that is higher tech level than that would be considered "Dangerous" in the hands of a normal human (Hence the issue with the Forever Knights as they are always seeking High level tech from other alien worlds). So I like to compare earth to say Tombstone Arizona circa 1870 or 1880. (In short Earth is a "Wild West style" planet with its low tech and the Plumbers here are like US Marshalls of the old West trying to bring criminals to justice.
But as for how many worlds the Plumbers are on? Well Space is pretty darn big, so we could be talking hundreds, thousands, even tens of thousands of worlds (Hey if Ben Can transform into 10,000 aliens (Refer back to Ben10,000) then that means there are at least that many worlds that theoretically could have plumbers on them.
Just my own observation on this.
